Lire la suite >Photodynamic therapy (PDT) is clinically used to treat different kind of cancers as well as age-related macular degeneration, acne... After excitation of a photo-activable molecule called the photosensitizer and energy transfer to oxygen, reactive oxygen species are produced leading to photooxidation reactions. One limitation of photodynamic therapy is the low selectivity of photosensitizers to tumor tissue compared to normal tissue. The synthesis of photosensitizers especially porphyrins or chlorins coupled with vectors specific of the membrane receptors over-expressed on neo-vessels or tumoral cells has become a center of interest.The poor outcome of primary malignant brain tumors derives from local invasion and local recurrence. Standard treatment of high-grade astrocytic tumors usually consists of cytoreductive surgery followed by radiation techniques and chemotherapy; however these tumor types usually recur despite treatments. In most cases, curative resection is not possible due to infiltrating growth of the tumor into normal brain parenchyma. In this unfavorable context, PDT appears as an innovative technology being investigated to fulfill the need for a targeted cancer treatment that may reduce recurrence and extend survival with minimal side effects. We develop peptide-targeted photosensitizers1,2,3 as well as peptide-targeted nanoparticles4,5,6 to improve the efficiency of PDT to treat glioblastoma.Ovarian cancer’s prognosis remains dire after primary therapy. 60% of women with epithelial ovarian cancer considered in remission will develop recurrent disease within five years. Special attention to undetected peritoneal metastasis during surgery is necessary as they are the main predictive factors of recurrences. Folate Receptor α shows promising prospects in targeting ovarian cancerous cells and intraperitoneal PDT could be a solution in addition to macroscopic cytoreductive surgery to treat peritoneal micrometastasis.We develop new folic acid targeted porphyrin7,8,9. The photodynamic activity of these targeted compounds are evaluated in vitro and in vivo.Lire moins >
